H3 Biomedicine is looking for a collaborative scientist to join our Biology team and support the development of novel oncology therapeutics. This individual is expected to run an innovative independent research program as well as collaborate with small molecule and drug discovery teams to generate novel oncology therapeutics. The candidate will participate in team efforts with scientists from Target Discovery and Genomics, Enzymology, Target Biology, Informatics and Medicinal Chemistry in the discovery and optimization of novel drug candidates. Use your scientific expertise to aid us in our structure-based drug design efforts. Your task will be the determination of high-resolution, three-dimensional structures of macromolecular drug targets (typically proteins), including crystallization and structure solution of medically important structures and target-ligand complexes. Principal Duties and Responsibilities: The successful candidate will lead the crystallography lab at H3 Biomedicine to support drug discovery program. Main responsibilities will include: · Ensure the delivery from recombinant protein design, protein expression, protein purification, crystallization, co-crystal structure determination, data collection and refinement as well as demonstrated skill in crystallization and use database tools to track crystallization trials. · Oversee build out, manage and maintain in house crystallography lab including hiring, setting up and maintaining infrastructure. · Staying scientifically current, in order to proactively identify, recommend and implement new technologies and capabilities. · Participate in our Oncology drug discovery efforts by carrying out crystallization, co-crystal structure determination, refinement, and on-site synchrotron data collection. · Work within a group that encompasses protein expression, purification, crystallography and NMR spectroscopy. He/she will collaborate with scientists throughout H3 Biomedicine. · Development of novel purification protocols as well as optimization of existing protein purification strategies and crystallizing proteins in the presence or absence of small molecule ligands
Job qualifications: · A PhD in Structural Biology, Biophysics, Biochemistry or a related discipline is required, in addition to relevant postdoctoral experience. The candidate will show evidence of sustained productivity and independent contributions in science, as demonstrated by a strong publication record and drug development experience in drug discovery · Familiarity with organic molecules coupled with the ability to effectively interface with synthetic and computational chemists to facilitate drug design is highly desired, as is previous RNA experience.as well as a track record of accomplishments demonstrating technical proficiency, structural thinking, and scientific creativity. · Hands on experience in solving X-ray crystal structures of proteins and protein complexes and familiarity with techniques such as ITC, surface plasmon resonance, ELISAs and/or immunoprecipitation to characterize proteins in support of structural biology is highly desired · Broad experience with a variety of biochemical methods for protein characterization (protein quantitation, protein modification strategies, cross linking, limited proteolysis). Familiarity with all techniques and hardware/software relevant to purifying protein constructs for crystallization and obtaining crystals. · Previous experience in pharmaceutical, biotechnology companies support drug discovery programs is a plus.
