Junior Specialist, Step I-II Genetics, Genomics, and Developmental Biology Division- Molecular & Cell Biology

To apply: https://aprecruit.berkeley.edu/apply/JPF01349

The nal lab in the Molecular & Cell Biology department at UC Berkeley is seeking two highly motivated individuals to carry out research that focuses on meiotic differentiation and its regulation. The research goals are 1) to understand how the degradative organelle known as the vacuole (lysosomes in mammals) undergoes remodeling during meiotic differentiation and 2) to understand the regulation of pathways that mediate transcription-interference of genes based on distal promoter usage in meiosis. We are looking for two motivated Junior Specialists with interests in tackling these questions using multidisciplinary approaches including molecular genetics, biochemistry and cell biology.

Execution and quantification of RNA extraction and Northern blotting experiments to analyze alternative transcript isoforms from yeast and mammalian cells.
Execution and quantification of chromatin immunoprecipitation experiments, followed by qPCR and NextGen sequencing for further data analysis.
Perform a suppressor selection strategy to identify factors involved in transcription-based gene silencing.
Execution and quantification of vacuolar immuno-isolation followed by mass spectrometry.
Perform FRAP experiments using TIRF microscopy.
Biotin-ligase based proximity labeling and immuno-isolation of proteins
Standard molecular biology and yeast genetic experiments for strain and plasmid construction relevant for the project
Maintenance of laboratory: maintaining adequate stocks; ordering supplies
Additional duties as assigned

Minimum/Basic Qualifications Required (By the time of application):
Bachelors Degree or equivalent degree in Molecular Cell Biology with emphasis on Genetics, or a related field.

Additional Required Qualifications (By start date):
The successful candidate will have at least 6 months of laboratory experience.
Skills in standard molecular biology including nucleic acid extraction, PCR, cloning, transformation, site directed mutagenesis, immunoprecipitation and Western blotting.
Skills in yeast tetrad dissection, fluorescence microscopy, knowledge in yeast genetics.
Skills in yeast protein extract preparation.
Skills in fluorescent microscopy.
